Efecto de la adición de tempol y catalasa al dilutor para la criopreservación de semen de alpaca

La criopreservación de semen en alpaca es una biotecnología reproductiva en desarrollo,
debido a la baja supervivencia de los espermatozoides tras el congelamiento. Es conocido
que el proceso de criopreservación involucra daños funcionales en los espermatozoides,
dado los diversos tipos de estrés a los que son sometidos, considerándose al estrés
oxidativo como uno de los más perjudiciales. El presente estudio busca reducir dicho
efecto empleando dos enzimas antioxidantes, Tempol y Catalasa, con el fin de optimizar
el proceso de criopreservación de semen en esta especie. Para realizar el proyecto se
trabajó con 13 eyaculados provenientes de 4 machos; cada eyaculado se dividió en 4
grupos: control (criopreservación estándar), antioxidante Tempol (adicionando un
análogo de Superóxido dismutasa), grupo Catalasa (adicionando catalasa al dilutor), y
grupo interacción Catalasa-Tempol. Las características seminales que se evaluaron antes,
durante y luego del congelamiento fueron: motilidad y producción de peróxidos lipídicos,
este último expresado en ng/mL de malondialdehído a través del método del ácido
tiobarbitúrico (TBA) modificado. El uso de los antioxidantes Tempol 1mM, Catalasa 50
ug/mL y su interacción como parte del dilutor para la criopreservación de
espermatozoides de alpaca no presentaron diferencias significativas sobre la motilidad
espermática (19.25 ± 11.11, 22.66 ± 21.88 y 14.82 ± 12.95 respectivamente) con respecto
al grupo control (19.04 ± 16.55). La producción de peróxidos lipídicos durante el proceso
de criopreservación de semen de alpaca no se ve reducida al utilizar antioxidantes
Tempol, Catalasa y su interacción (293.2 ng/mL, 240.3 ng/mL y 169.4 ng/mL
respectivamente) con respecto al grupo control (338.4 ng/mL). En conclusión, el uso de
antioxidantes no mejora la motilidad post descongelamiento ni reduce la producción de
peróxidos lipídicos en los espermatozoides de alpaca.
